Resumo: A alelopatia é um processo pelo qual produtos do metabolismo secundário de um vegetal são liberados, interferindo no desenvolvimento de outras plantas relativamente próximas. O interesse na exploração de compostos do metabolismo secundário é visto como alternativa estratégica na agricultura, inclusive para o controle de plantas invasoras. Apesar do aumento nas últimas décadas nas pesquisas por plantas com potencial alelopático, poucos estudos foram realizados sobre o perfil químico ou a atividade biológica de espécies nativas. A espécie Ocotea puberula (Rich.) Nees (Lauraceae), conhecida como guaicá, é uma espécie medicinal endêmica da Floresta Ombrófila Mista, utilizada principalmente por índios do Paraná e Santa Catarina. O objetivo desse trabalho foi avaliar a atividade alelopática de extratos CHCl3 e AcOEt das folhas de O. puberula sobre a germinção de Lactuca sativa L (alface). O extrato etanólico foi obtido a partir do material seco e moído. O extrato bruto foi solubilizado em etanol e acidificado (fração hidroalcoolica). A fração C6H14 foi obtida por extração líquido-líquido da fração hidroalcoolica, mesmo procedimento utilizado para extração da fração AcOEt. A fração hidroalcoolica foi basificada e extraída com CHCl3. O perfil químico da frações obtidas foi analisado por Cromatografia em Camada Delgada (CCD) utilizando-se reveladores específicos. A análise por CCD confirmou a presença de alcalóides como compostos majoritários nas frações AcOEt e CHCl3. As frações foram avaliadas quanto ao potencial alelopático, nas concentrações de 0,8 gmL-1, 0,4 gmL-1 e 0,2 gmL-1. Os testes de germinação foram realizados em câmara germinativas à 25ºC, em placas de Petri contendo 25 sementes de alface com quatro repetições. Para o teste de crescimento foram utilizadas 10 plântulas por placa de Petri prégerminadas seguindo os mesmos procedimentos anteriores. Os dados foram submetidos à ANOVA e comparados pelo teste de Scoot-Knoot (p>0,05). Os parâmetros germinativos analisados foram porcentagem de germinação (%G) e o índice de velocidade de germinação (IVG). Os resultados indicaram alterações significativas na %G no IVG e no crescimento radicular (CR) das sementes de alface. Todas as concentrações da CHCl3 reduziram significativamente os parâmetros germinativos avaliados, sendo que a concentração de 0,8gmL-1 diminuiu 50% a %G e 80% o IVG e 74% o CR, quando comparados com o controle. Já as AcOEt não interferiram na %G, porém todas as concentrações aplicadas reduziram significativamente o IVG e o CR. Muitas vezes o efeito alelopático não se manifesta sobre a %G, mas sobre o IVG das sementes. As frações AcOEt e CHCl3, extraídas das folhas de O. puberula, apresentaram influência alelopática sobre a germinação de alface. No entanto a CHCl3 mostrou-se mais efetiva, provocando uma redução acentuada na %G no CR e no tempo de germinação das sementes.
Abstract: Allelopathy is a process whereby the compounds of the secondary metabolism of a plant are released, interfering in the growth of other relatively close plants. The interest in the exploration of compounds from the secondary metabolism is seen as a strategic alternative in agriculture, even for the control of invasive plants. Despite the increase in the last decades on the research by plants with allelopathic potential, few studies have been conducted on the chemical profile or the biological activity of native species. Ocotea puberula (Rich.) Nees (Laureaceae), known as guaicá, is a medicinal species endemic to the Ombrophilous Mixed Forest, mainly used by Brazilian indians on Paraná and Santa Catarina states. The objective of this work was to measure the allelopathic activity of extracts CHCl3 and AcOEt from the leaves of O. puberula on the germination of Lactuca sativa L. (lettuce). The ethanolic extract was obtained from the dried and milled leaves. The crude extract was solubilized in ethanol and acidified (hydroalcoholic fraction). The C6H14 fraction was obtained by liquid-liquid extraction of the hydroalcoholic fraction, the same procedure was used to extract the AcOEt fraction. The hydroalcoholic fraction was basified and extracted with CHCl3. The chemical profile of the obtained fractions was analyzed by Thin Layer Chromatography (TLC) using specific revelators. TLC analysis confirmed the presence of alkaloids as majority compounds in AcOEt and CHCl3 fractions. The fractions were evaluated for allelopathic potential on lettuce cypselas at 0.8 gmL-1, 0.4 gmL-1 and 0.2 gmL-1. Germination tests were carried out in a growth chambers at 25ºC with Petri dishes containing 25 lettuce cypselas with four replicates. For the growth test, were used ten pre-germinated seedlings per petri dish, we followed the same procedures as above. The data were submitted to ANOVA and compared by the Scoot-Knoot test (p> 0.05). The germination parameters analyzed were: germination percentage (% G) and germination speed index (IVG). The results indicated significant changes in the %G and the IVG and root growth (CR) of the lettuce seeds. All CHCl3 concentrations reduced significantly the germinative parameters evaluated, and the concentration of 0.8gmL-1 decreased 50% to %G and 80% the IVG, and 74% the CR. when compared to the control. The AcOEt did not interfere with the %G, but all the concentrations applied significantly reduced the IVG and CR. Sometimes the allelopathic effect does not manifest on the % G, but only in the IVG of the seeds. The AcOEt and CHCl3 fractions, extracted from the leaves of O. puberula, revealed allelopathic influence on lettuce germination. However, CHCl3 showed to be more effective, with marked reduction in the %G and CR and in the germination time of the seeds.
